Abstract:
The main question which lead us to do such project is that whether the biodiversity of soil differs from that of different places .All of us suppose that the soil biodiversity here in Suzhou Industrial Park (SIP) is worse than that of original soil. To test this hypothesis, we would like to use two different methods to get DNA sequence from soil. First of all, we will collect soil and use insect phototaxis and Berlese traps. We will use the method of Chelex DNA Isolation to get the DNA of insects. Then, we will use PCR to amplify the COI DNA region. After that, the section will be sequenced to get. Finally, we will use DNA Subway to identify species to classify and summarize the obtained data according to soil depth. In addition, we’d like to get microbial DNA directly from the soil sample by 16S Extraction. These barcodes will be sequenced by nanopore technology, and we will determine microbial diversity. Over all, we compare our data to data of Canadian soil from another study.
